2 vii Abstract The osseointegration of titanium implants is known to be significantly influenced by their surface roughness. This surface property has a significant effect on cell behaviour, such as adhesion, proliferation and differentiation. In order to investigate the effect of roughness on cell attachment, roughness gradients offer an ideal, high- throughput solution [1]. While previous studies [2] have shown a higher osteoblast proliferation on micrometer- sized roughness compared to flat references, nanometer- scale roughness was shown to slow down cell proliferation [3, 4]. In order to understand the effect of roughness scale on the behavior of osteoblasts, new particle- size gradients have been developed. In this thesis, three new techniques have been investigated: The oxidation of organic particles through UV illumination on a titanium oxide layer was used to generate particle- height gradients. Such gradients were combined with a particle- density gradient to generate a 2- dimensional particle- height vs. particle- density gradient. Separation and deposition of a polydisperse particle suspension was performed by means of an electric field in an acrylamide gel. The preparation of such polydisperse silica- particle suspensions required a study of the kinetics of the well- known Stöber synthesis, and an adaptation of the synthesis protocol.

3 viii Abstract Silver or gold layers were deposited with varying thicknesses on silicon wafers, followed by solid- state dewetting at 600 C, to fabricate a particle- size gradient, with features ranging between 300 nm and 5 nm. All gradients were extensively characterized with AFM and SEM. A replication technique was also adapted for organic particles on a silicon substrate to ensure high- fidelity epoxy replicas. These replicas were further coated with titanium to mimic a bone implant. The particle- height vs. particle- density gradients were further used for cell- culture tests, and the proliferation of osteoblasts was studied. It was shown that the higher the particle density, the faster the osteoblasts proliferated on the substrate. The particle height did not play any significant role in cell proliferation, but the shape of the cells was found to be different. It was observed that for lower particle heights, the cells adopt a more spread conformation on the substrate, increasing the cell area coverage. The gold dewetted gradient was further backfilled with titanium and template- stripped for the generation of ultra- flat bimetallic samples with gold islands embedded in titanium. Such samples were used as standards for surface- chemical sensitive techniques such as ToF- SIMS and Auger imaging spectroscopy, in order to determine the lateral resolution of the instruments. The gold and silver dewetting technique was also used for the generation of silicon substrates with varying porosity with metal- assisted chemical etching. The size of the metal islands could be varied, leading to varying pore sizes in the final silicon wafer. Finally, the afore- mentioned replication technique [5] was further developed through the use of a lost- mold technique for the preparation of micro- featured ceramics. 5 xi Résumé L ostéointégration des implants en titane est connue pour être fortement influencée par leur rugosité de surface qui affecte de façon significative le comportement cellulaire, tel que l adhésion, la prolifération et la différentiation. Pour étudier l effet de la rugosité sur des cellules, les gradients de rugosités sont une méthode rapide et pertinente [1]. Une étude précédente [2] a montré que la prolifération des ostéoblastes est plus rapide sur une rugosité micrométrique que sur une référence plate, alors qu une rugosité nanométrique a prouvé une diminution de la prolifération cellulaire [3, 4]. Pour comprendre l effet de l échelle de rugosité sur le comportement des ostéoblastes, de nouveaux gradients de taille de particules ont été étudiés. Trois nouvelles techniques de fabrication de ces gradients sont développées dans cette thèse : L oxydation de particules organiques exposées à des UV sur une couche d oxyde de titane a permis de produire des gradients de hauteur de particules. Ces gradients ont été combinés avec un gradient de densité de particules pour créer des gradients selon 2 dimensions : hauteur de particules et densité de particules. La séparation et la dépose d une suspension polydisperse de particules a été réalisée par l action d un champ électrique dans un gel d acrylamide. La préparation de ces suspensions polydisperses de particules de silice a

6 xii Résumé nécessité une étude approfondie de la cinétique de réaction de la synthèse Stöber, ainsi qu une adaptation du protocole de synthèse. Le dépôt de couches d argent ou d or avec une variation d épaisseur sur des wafers de silicium, suivis d un démouillage sous forme solide à 600 C, a permis la fabrication d un gradient de taille de particules de dimensions comprises entre 5 nm et 300 nm. Tous les gradients ont été caractérisés avec un microscope à force atomique ou un microscope électronique à balayage. Une technique de réplication a été adaptée pour les particules organiques sur un substrat de silicium, pour assurer la fidélité des répliques en époxy. Ces répliques ont ensuite été recouvertes d une fine couche de titane pour imiter un implant osseux. Le gradient à deux dimensions (hauteur de particules et densité de particules) a été utilisé pour étudier la prolifération des ostéoblastes. Cela a permis de démontrer que plus la densité de particules est élevée, plus les ostéoblastes prolifèrent vite sur le substrat. La hauteur des particules n a pas montré d effet significatif dans la prolifération cellulaire, mais a modifié la forme des cellules. Il a été observé que moins les particules sont hautes, plus les cellules s étalent sur le substrat, augmentant ainsi l aire couverte par les cellules. Ces méthodes de fabrications de gradients nous ont incités à rechercher d autres applications : Le gradient d or démouillé a été rempli de titane et séparé de son support pour générer des échantillons bimétalliques ultra plats présentant des ilots d or incorporés dans du titane. Ces échantillons ont été utilisés pour estimer la résolution latérale d instruments de mesures sensibles à différentes chimies de surfaces tels que le ToF- SIMS et l imagerie par spectroscopie Auger.

7 Résumé xiii La technique de démouillage d or et d argent a aussi été utilisée pour créer des structures ressemblant à des membranes, par attaque chimique à l aide de métaux. La taille des ilots de métal a été modifiée et cela a changé la taille finale des pores obtenus dans le wafer de silicium. Finalement, la technique de réplication en alumine [5] a été développée plus avant en utilisant une technique de moule perdu pour la préparation de céramiques micro- structurées. Des structures de 2 µm ont été réalisées avec un rapport d aspect de 2.
