2 Summary Lv. Summary For the amelioration of the Standard methods used in cattle breeding and research, an improved collaboration between the breeding biology and the reproduetive biology is needed. The combination of these exciting fields of research should lead to the developmentof new dairy and beef cattle breeding methods. Such collaboration will increase the accuracy of the predicted genetic value of animals, reducing the generation interval and increasing the genetic improvement of animals achieved in each generation. To enhance these steps, this research work focused on the transfer of in vitro produced embryos previously biopsied and genotyped. For the in vitro production, bovine ovaries were obtained from slaughtered Brown Swiss and Simmental cows. Recovered cumulus-enclosed ooeytes were matured and fertilized and the ooeytes were eultured in vitro under Standard conditions. Several Red Holstein, Holstein, Brown Swiss and Limousin bulls were used for the in vitro production of embryos. Fertilization for the assessment and control of the transmission of 1/29 centromere fusion was performed using sperm from a Brown Swiss bull heterozygote for the centric fusion with proven natural fertility records. For the amplification of DNA of the embryos, half-embryos and biopsied blastomere were used following a preamplification Polymerase chain reaction (PEP-PCR). Of the 450 füll embryos analyzed by PEP-PCR and the subsequent amplification of microsatellites, 371 (80%) showed the presence of markers used to test the amplificationpattern of the DNA with this technique. The DNA of the parent animals was extracted from muscle or sperm and the genotypes were determined. Single trait genes and microsatellites were amplified to analyze the correlated genotype and alleles of the embryo. An ABI PRISM 377

3 Summary Y_ DNA Sequencer in combination with GeneScan Analysis Software or agarose gel electrophoresis was used to determine the size of the amplified products. Typing of six microsatellite markers (AGLA17, BM6438 and TGLA49 on chromosome 1 and BM4602, BMC2228 and BMS1857 on chromosome29) for 1/29 Robertsonian translocation in cattle resulted in haplotype combinations (222, 262, 116, 113, 173 and 159, respectively) which segregate with the 1/29 Robertsonian translocation.the blastocyst rate obtained at Day 8 was 39%, a result similar or even slightly higher than with the in-housecontrol bulls. None of the cows had the six alleles linked to this centromere fusion. From the 93 embryos obtained in this study, 48 (51%) showed the defined haplotype combination segregated with the 1/29 Robertsonian translocation. The results showed that a 1:1 ratio (x2=0.097, p»0.05) exists for fertilization and this translocation did not impair the in vitro development of the blastocysts up to Day 8. The developed minimal method of biopsied embryo revealed a blastocyst rate of 36%. Of the 72 embryos biopsied with the rolling and extrusion method developed during this work, 26 embryos were produced in vitro after achievementof the embryo biopsy. ET of biopsied and frozen embryo were done with 13 embryos, resulting in the birth of two calves, thus resulting in a 15% pregnancy rate. The genotypes of the tested calves were in accordancewith the genotype of their respective mothers. The metaphase analysis of the two born calves, one femaie and one male, showed the normal set of 60 chromosomesconfirming the microsatellite analyses. The parental relation was assessed by parentage control with microsatellites. Both, the blastomere analyzed and the calves show a consistent inheritance pattern with the bull and the cows considered as mother. The combination of reproduetive technology with genomic analysis methods should in the future lead to improved breeding values. The selection of embryo

5 Resume _YÜ_ Resume Afin d'ameliorer les methodes appliquees dans le domaine de la recherche animale et de l'elevage bovin, une collaboration accrue entre la biologie de l'elevage et la biologie de la reproduction est necessaire. La combinaison de ces deux domaines d'application passionnants devant aider ä ameliorer les methodes de management des ressources genetiques dans le domaine de la production bovine laitiere et bouchere. Cette collaboration amenera une efficacite accrue lors de la prediction de la valeur genetique des animaux, tout en reduisant les intervalles de generations et en augmentant le gain genetique obtenu par generation. Dans ce but, le travail presente est cible dans le domaine du transfert d'embryons produit in vitro prealablement biopsies et analyses genetiquement. Pour la production in vitro d'embryons, les ovaires bovins de vaches et genisse des races Simmental et Brown Swiss ont ete preleves ä l'abattoir. Le Processus de maturation, fertilisation et culture des complexes ovocytes cumulus a ete accompli in vitro sous conditions Standards. Differentstaureaux des races Red Holstein, Holstein, Brown Swiss et Limousin ont ete utilises pour la production in vitro des embryons. La fecondation des ovocytes destines ä l'etude et au contröle de la transmission de la fusion translocative robertsonnienne des centromeres 1/29 a ete realisee au moyen du sperme d'un taureau Brown Swiss, porteur heterozygote de cette fusion de centromeres. La fecondite de ce taureau avait ete testee prealablement en monte naturelle. Des embryons, demis embryons et blastomeres biopsies ont ete utilises afin d'effectuer une amplification de l'adn par amplification en chaine par Polymerase (PCR) consecutivement ä une preamplification en chaine par Polymerase (PEP-PCR). Des 450 embryons entiers analyses par PEP-PCR et microsatellites, 371 embryons (80%) ont montres une amplification de

6 Resume VIII marqueursgenetiques positive. Le profil genetique de l'adn des parents, extrait de muscles et du sperme, a egalementete determine. Plusieurs genes ont ete utilises pour l'analyse des genotypes et alleles des embryons. La longueur des sequences des produits d'amplification ä ete analysee avec un sequenceur ABI PRISM 377 DNA et le Programme correspondant (GeneScan Analysis Software) ou par electrophorese sur gel. Pour la typisation de la fusion des centromeres 1/29, l'amplification de six marqueurs genetiques ou microsatellites ont ete utilisee (AGLA17, BM6438 and TGLA49 sur le chromosome 1 and BM4602, BMC2228 and BMS1857 sur le chromosome 29). La typisation aboutit ä la determination des combinaisonsd'haplotype (222, 262, 116, 113, 173 and 159, respectivement) segregant avec la fusion translocative robertsonnienne 1/29. Le taux de blastocystes obtenu ä J8 est de 39%, un resultat similaire ou meme legerement superieur aux taux observes avec les groupes de contröle. Aucune des vaches anaiysees n'etait porteuse de la fusion des centromeres 1/29. L'analyse des 93 embryons obtenus lors de cette etude, montre que 48 embryons (51%) sont porteurs de la combinaison des haplotypes segregant avec la fusion des centromeres 1/29. Par ce resultat, il est demontre d'une part, qu'un ratio de 1:1 (x2=0.097, p»0.05) est obtenu lors de la fecondation et, d'autre part que la translocation n'influence pas le developpementin vitro des blastocystes jusqu'ä J8. Avec la methode de biopsie appliquee, un taux de blastocystes J8 de 36% a ete obtenu. Des 72 embryons biopsies selon la methode dite de «roulage et extrusion» developpeedurant ce travail, 26 embryons ont ete produits in vitro consecutivement ä l'extraction d'un ou plusieurs blastomeres. Par la suite 13 embryons biopsies et cryogenises ont ete transplant.es. obtenues correspondent ä un taux de gestation de 15%. La determination des genotypes des veaux montre une concordance avec le genotype Les deux naissances des meres presomptives. Par l'analyse des metaphases des 2 veaux nes, un male et une femelle, il a pu etre etabli qu'ils ont tout deux 60 chromosomes, corroborant ainsi les analyses faites au moyen des microsatellites. La determination du pedigree a ete effectuee par genotypisation par microsatellites. Tant l'analyse

7 Resume IX des blastomeres biopsies que l'analysedes veaux ont confirmes les filiations du taureau et des meres considerees. La mise en reseau et combinaison des techniques de reproduction avec les methodes d'analyse genomique laisse esperer une amelioration concrete des pratiques des methodes d'elevage actuelles. La preselection genetique d'embryons produits in vitro est une technique delicate ä implementer, mais utile aux progres constants vises en zootechnie par la mise en reseau des domaines fortement lies entre eux que sont la biologie de l'elevage, l'etude de la reproduction et la genetique animale.
