18 Proposal abstract: The main objective of this 4 years IP is the development, up to clinical phase I, of a specific HIV microbicide for the prevention of sexual transmission/acquisition of HIV, based on the interruption of viral replication at the mucosal level. Some members of a new class of non-nucleoside reverse transcriptase inhibitors (NNRTIs), the DABOs, proved capable of irreversibly knocking out HIV-1 replication. This property correlates with their ability to tightly bind to the HIV-1 Reverse Transcriptase. The lead compound of this series of tight binding NNRTIs is MC1220. The global preclinical and pharmaceutical workflow is organised in 4 sub projects to permit very rapid access to formulation screening and challenge efficacy model in the macaque in 2 centres, 1 in Europe, 1 in Africa. Finally the clinical phase I trial (tolerability studies in women) will involve HIV medical teams very much implicated towards development of clinical trial and treatment networks in Africa. 11 partners, from academy to industry will contribute to the project & focus both on respect of timing and achievement of expected objectives, as well as to widespread scientific results and economical benefits to achieve all scientific and regulatory requirements to go to phase II clinical trial programmes within EDCTP or Alliance for microbicide for example. 18

Ensure that the potential properties Reduce slightly the budget (travel, personnel costs ) Provide information on how the consortium will resolve conflicting issues especially when members cannot reach consensus Reinforce the training component in all aspects of research (basic and clinical) with a special emphasis on training representatives from DCs at all levels of their scientific career Start identifying partners for conducting clinical trials in DCs as early as possible Clarify whether the new share-holder will continue its commitment to the project 22

23 1. Relevance (Threshold 3/5; Weight 1) Highly relevant to the objectives of the work programme.(4.5 / 5) Each of the proposed areas will contribute to the empowerment of women worldwide. The consortium members come from 5 different European countries (France, Italy, Germany, Denmark and Greece) as well of developing country (Gabon) and involves a SME Idenix. Training component is associated with technology transfer but is limited to Gabon. 23

Potential impact (Threshold 3/5; Weight 1) (4.5/ 5) A NNRTI based topical microbicide for use by women during intercourse for the prevention of HIV could have an enormous impact on the heterosexual spread of HIV. Some of the partners involved in the project have a proven track record in the academia-industry transfer of technology in the field of antiretroviral treatment. The SME, Idenix, has already set up developing processes ensuring the rapid scale up and production for preclinical and clinical studies. The panel noted however that Idenix is now taken over by Novartis. The consortium brings together highly specialised scientists with a clear path to success that could put Europe at the forefront of microbicide development. 24

S&T excellence (Threshold 4/5; Weight 1) 4.5/5 This is a very strong project due to its very clear and focused set of objectives; The path which needs to be followed to meet those objectives is also very well described. Several elements in the proposal could result in research innovation in this field. The major areas of contribution beyond the stateof-the-art lie on the following areas. Cleary not all aspects of drug development can be scientifically exciting. The investigators recognize that aspect of the developmental process and have those activities performed under contracts. Their implementation plan is very well defined and their risk assessment and contingency plan is also well thought out. 25

26 4. Quality of the consortium (Threshold 3/5; Weight 1) 5/5 The consortium is of outstanding quality. Each partner has excellent qualifications and proven records in the field. Individual role has been clearly defined. The complementarity between various teams is well established with no overlaps. Each contribution is essential to the final goal of developing a safe and effective microbicide. It appears the industry will play a key role that needs to be counterbalanced by input from public institutions in a real collaboration. 26

27 5. Quality of the management (Threshold 3/5; Weight 1) 4.5/5 The organisation structure of the consortium is of high quality and has been well described. It is very adequate to manage this manageable project and will allow close integration of individual projects. It includes strategic, executive and implementation level of activities that will provide at the same time cohesion and flexibility. 27

28 6. Mobilisation of the resources (Threshold 3/5; Weight 1) 4/5 There is minimum critical mass for the smooth running of the projects within the required budget. It would appear that most of the actual testing will be carried out in Germany, perhaps more consideration should be given to equipment and testing in Gabon enabling this country to become a reference centre. Some cuts to the budget could be done on travelling, salaries, and overheads. A clear presentation of the projected costs for each of the proposed subprojects could be clearly presented. 28

29 EVALUATION ETHIQUE The project aims to produce a topical microbicide, up to clinical phase I, for vaginal use to prevent sexual transmission/ acquisition based on the interruption of viral replication at the mucosal level. Ethical Issues Raised Applicants made a great effort to address ethical questions. The proposal is very conscious of potential ethical issues. - use of human blood and tissues; - use of non-human primates for validation; - employment of adult healthy volunteers; - clinical trials involving HIV+ and HIV- women; - plans for clinical trials in Africa; - empowerment of women; 29

Oui, si le programme de travail du 6ème PCRD colle avec sa stratégie Si le projet ne porte pas sur des axes à risque pour l entreprise Si elle peut dégager des ressources dès la rédaction du projet pour être acteur, voire moteur du projet Si elle isole du projet son savoir faire clé qu elle ne souhaite pas divulguer Si elle se crée une place «démocratique» au sein des structures de management et décision du projet, surtout si des grands groupes y participent. Cependant 31

32 Concernant la sous-traitance Remarque 1: La Commission tolère la sous-traitance, si elle est justifiée. Pensez à joindre vos appels d offre, les modalités de choix du soustraitant et à démontrer que vous n avez pas pu l intégrer en tant que partenaire. Remarque 2: Si vous avez un profil de sous-traitant, vous aurez du mal à rentrer dans le 6ème PCRD Remarque 3 : Certaines tâches ne peuvent être effectuées que par de sociétés de service spécialisées (pas d inventivité particulière mais des garanties de qualité, et de conformité réglementaires GLP, GMP).
