2 iii Abstract The growing demand of biopharmaceutical products is boosting the market for therapeutic recombinant proteins (r-proteins). More than half of the 140 r-proteins that have gained approval for human therapeutic use are manufactured in mammalian cells that make possible complex post-translational modifications, like glycosylation. In the industry, r-proteins are manufactured by stable gene expression (SGE) following Good Manufacturing Practice (GMP) standards. Transient gene expression (TGE) in mammalian cells is an alternative method for the expression of r-proteins whose main advantages are rapidity and flexibility. TGE involves the expression of the protein of interest from plasmids, which are transfected into the cells with a non-viral DNA transfecting agent, usually polyethylenimine (PEI). DNA is transcribed from the plasmids without the latter having been integrated into the host genome. Despite the recent improvements in r-protein titers by TGE and the scale up of this method to the 100-L scale, TGE remains so far a tool for protein production for research purposes. Therefore, to determine if TGE can be used for manufacturing therapeutic r-protein following GMP standards, different features related to reproducibility and product quality from TGE of a recombinant anti-rhesus D immunoglobulin G in HEK-293E cells using linear PEI were characterized. To test the effects of the size distribution and chemical structure of PEI on TGE, different commercially available PEIs were characterized and their impact on transfection and r-protein expression were assessed. The results showed that both the molecular weight distribution and the chemical structure of the PEI had an effect on TGE. However, the small variations in size distribution and structure between the batches of PEI from the same supplier did not affect significantly the transfection and r-protein production by TGE. Then, the fate of PEI and plasmid DNA in cell culture over time and their removal from the final r-protein during purification were determined and measured. Most of the PEI and pdna remained in the cell culture medium after transfection. However, both PEI and plasmid DNA, which are additional components in TGE compared to processes based on recombinant cell lines, could be reduced to a concentration below the limit of detection of the assays after Protein A affinity purification of the antibody. To test the reproducibility of a TGE process, 10 batches of transfected cells were run in 500-mL orbitally shaken bottles. The cell specific productivity of the antibody was 20.2 ± 2.6 pg.cell 1.day 1 on average for the 10 batches. The purity and size consistency of the recombinant antibody produced in those 10 batches was demonstrated by gel electrophoresis and size exclusion chromatography. Then, the glycosylation profile of the antibody

3 iv produced by TGE in HEK-293E cells was determined by mass spectrometry and was reproducible over the 10 batches. Moreover, it was similar to the glycosylation profile of the antibody produced by 3 stable HEK-293E cell lines. Finally, as a proof of concept, TGE was applied for the development of a vaccine against the respiratory syncytial virus. The fusion protein of the respiratory syncytial virus was produced by TGE and used as an antigen for the development of a recombinant vaccine. TGE enabled the rapid evaluation of the candidate vaccine in animal trials. Overall, the results of this study suggest that TGE is a reliable and reproducible method for manufacturing r-proteins of a consistent quality, provided that some particular features, such as reagents quality and process parameters, are well defined and controlled. Since TGE makes possible the rapid production of virtually any protein, even toxic, it could be used to provide GMP approved biopharmaceuticals for clinical trials in a short time, reducing development costs of biological therapeutic products. 4 v Résumé La demande croissante des produits biopharmaceutiques a accéléré l expansion du marché des protéines recombinantes (r-protéines) à but thérapeutique. Plus de la moitié des 140 r-protéines homologuées par les autorités compétentes en vue d une utilisation thérapeutique sont fabriquées par des lignées cellulaires mammifères pouvant effectuer des modifications post-traductionnelles complexes, telle que la glycosylation. Dans l industrie, les r-protéines sont fabriquées par expression génétique stable (EGS) conformément aux Bonnes Pratiques de Fabrication (BPF). L expression génétique transitoire (EGT) dans les cellules mammifères est une méthode alternative permettant la production de r-protéines; elle présente deux avantages majeurs : la rapidité et la flexibilité. L EGT consiste en l expression d une protéine d intérêt à partir de plasmides, qui sont transfectés dans la cellule à l aide d un agent de livraison non viral de l ADN, d ordinaire le polyethylèneimine (PEI). L ADN est transcrit à partir du plasmide sans que ce dernier ait été intégré dans le génome de la cellule hôte. Malgré les améliorations récentes apportées aux titres de r-protéines par EGT et la démonstration de l EGT pour des volumes de 100 L, l EGT reste un outil pour produire des protéines à des fins de recherche. En conséquence, pour déterminer si l EGT peut être utilisée pour la production de r-protéines thérapeutiques conformément aux BPF, plusieurs aspects liées à la reproducibilité ainsi qu à la qualité du produit provenant de l EGT d une immunoglobuline G recombinante anti-rhesus D dans des cellules HEK-293E à l aide de PEI linéaire ont été caractérisés. Afin de tester les effets de la distribution de poids moléculaire et de la structure chimique du PEI sur l EGT, plusieurs PEI disponibles dans le commerce ont été caractérisés, et leur impact sur la transfection et la production de r-protéines a été évalué. Les résultats ont montré que la distribution de poids moléculaire ainsi que la structure chimique du PEI ont influencé l EGT. Toutefois, de légères variations de distribution de taille et de composition entre les différents lots de PEI provenant d un même fournisseur n ont pas eu d effet significatif sur la transfection et la production de r-protéines par EGT. Ensuite, ce qui advient du PEI et de l ADN plasmidique dans la culture cellulaire au fil du temps ainsi que leur élimination de la r-protéine finale lors de la purification ont été recherchés et mesurés. La plupart du PEI et de l ADN plasmidique est restée dans le milieu de culture après la transfection. Cependant, les résultats montrent que le PEI et l ADN plasmidique, qui sont des composants supplémentaires dans l EGT par rapport aux procédés basés sur des lignées cellulaires stables, ont pu être éliminés et réduits à des concentrations inférieures aux limites

5 vi de détection des techniques de mesure, après avoir procédé à une purification de l anticorps par affinité à la Protéine A. Puis, afin d évaluer la reproducibilité d un procédé d EGT, 10 lots de cellules transfectées ont été répétés dans des bouteilles de 500 ml agitées de façon orbitale. Une productivité cellulaire spécifique de l anticorps s élevant en moyenne à 20,2 ± 2,6 pg.cellule 1.jour 1 a été mesurée pour les 10 lots. La constance de la pureté et du poids moléculaire de l anticorps produit dans ces 10 lots a été vérifiée par gel d électrophorèse et chromatographie d exclusion stérique. Ensuite, le profil de glycosylation de l anticorps produit par EGT dans les cellules HEK-293E a été déterminé par spectrométrie de masse et était reproductible dans les 10 lots. En outre, il était similaire au profil de glycosylation de l anticorps produit par 3 lignées cellulaires stables recombinantes HEK-293E. Enfin, afin d en démontrer la faisabilité, l EGT a été appliquée au développement d un vaccin contre le virus respiratoire syncytial. La protéine de fusion du virus respiratoire syncytial a été exprimée par EGT et utilisée en tant qu antigène pour le développement d un vaccin recombinant. Ainsi, l EGT a permis d évaluer rapidement le vaccin lors de tests précliniques. Dans l ensemble, les résultats de ce travail de recherche indiquent que l EGT est une méthode fiable et reproductible pour fabriquer des r-protéines de qualité constante, pour autant que certains aspects spécifiques, tels que la qualité des réactifs et les paramètres du procédé, soient définis et contrôlés. L EGT, qui permet la production rapide de pratiquement n importe quelle protéine, même toxique, pourrait être utilisée, conformément aux BPF, pour la fabrication de r-protéines destinées aux tests cliniques de produits biopharmaceutiques, ce qui réduirait le temps et les coûts nécessaires à leur développement.
