3 In memoriam Radovan Popovic Un directeur de recherche canadien reconnu en photosynthèse moléculaire appliquée à l environnement, un mentor exceptionnel, un collègue et ami très apprécié par beaucoup de personnes. Au cours de sa longue carrière, Radovan a fortement contribué à l'utilisation de la fluorescence de la chlorophylle en toxicologie environnementale afin de mieux comprendre les mécanismes d action de nombreux polluants aquatiques. 3

Laboratory of Environmental Toxicology, LABTOX Department of Sanitary and Environmental Engineering, Federal University of Santa Catarina - Campus Universitário Trindade - Florianopolis, SC - CEP Brazil Les réseaux de recherche coopérative ont été un excellent moyen de catalyser le développement d'études, de formation des ressources humaines scientifiques et de développer les relations entre les universités et les entreprises. L'association de groupes de recherche qui complètent techniquement, pour développer un produit, c'est un excellent moyen d'optimiser le temps et les ressources. Bien sûr, les relations humaines est fondamentale pour la création et le développement d'une coopération rendements. Le développement des coopératives à travers l'articulation entre les institutions scientifiques et les entreprises peuvent accélérer le processus de la connaissance des risques et des impacts potentiels des nouvelles technologies basées sur les nanosciences, la promotion de l'avancement des connaissances scientifiques et en toute sécurité technologique. En ce sens, les groupes qui composent notre réseau coopératif de recherche ont des caractéristiques différentes, mais complémentaires. Les compétences individuelles de chaque institution est fondamentale pour le développement de la recherche, l'échange d'informations scientifiques et la formation des ressources humaines. Ainsi, la capacité technique et de bonnes relations humaines entre les chercheurs, UFSC (Department of Environmental Engineering et Sanitátria), UQAM (Département de chimie et biochimie), Université de Bordeaux II (Laboratoire de toxicologie), Rice University (Department of Environmental Engineering), UNIFESP (Département des sciences exactes et de la Terre - Département des Sciences Pharmaceutiques), UNIVILLE (Laboratoire d'écotoxicologie) avec des fonds du CNPq, mis en place un réseau coopératif de recherche en nanotoxicologie, appliqué au secteur des revêtements (peintures) et du pétrole. Les premiers résultats seront présentés, basée sur des études avec des nanoparticules métalliques à l'aide de différents modèles toxicologiques, comprenant la toxicologie globale (études de toxicité aiguë et chronique) et spécifiques (génotoxicité, l'épigénétique et oxydatif). Their extensive use can contaminate aquatic ecosystems. However, the toxicological effects of this NP in the environment are poorly known. In this study, we evaluated the toxicity and oxidative stress induced by CuO NP on Chlamydomonas reinhardtii using several toxicological assays. CuO NP was found to induce growth inhibition and a significant decrease in carotenoids levels. Evaluation of esterase activity demonstrates a decrease in cell metabolism activity with the increase of CuO NP concentration. The CuO NP induced an increase of reactive species (RS) level and lipid peroxidation of cellular membranes. Investigation of CuO NP uptake showed the presence of NP into the C. reinhardtii cells in different sites of the cell and, biomarkers of enzymatic antioxidants showed a change of activity after CuO NP exposition. In conclusion, C. reinhardtii was shown to be sensitive to the presence of CuO NP in their environment and CuO NP treatments induced a toxic response from 0.1 mg L -1 after 72h of treatment. Currently, the use and applicability of copper oxide nanoparticles has been increased considerably, mainly on science and industry field. However, the studies developed to assess health risks and environmental impacts are scare. Therefore, the present study intent to evaluate the toxicological effects of Copper Oxide Nanoparticles (CuO NP). The methodology used is based on standards procedures developed by ISO DIS (2004) and the OECD (2002), however, using the duckweed Landoltia punctata, as test organism. Also, the genotoxic effects will be evaluated by micronucleus assay. The CuO NP was characterized using transmission electron microscopy and zeta potential analysis aiming to evaluate the nanoparticles size and form, in solution, and samples stability. The duckweeds were taken from natural ponds and adapted to laboratory environment to maintenance of pre-culture. The concentrations of NP CuO samples chosen to the test were 0.4, 0.2 and 0.1 g.l -1. The results of toxicity will be taken based on the assessment of plant growth rate and micronuclei frequency. For now, the results obtained show that the duckweed species Landoltia punctata meeting the growth rate requirements, according to reference standard, and the pre-culture is able to be used in test. As results, is expected to identify the concentration range of NP CuO that cause disturbs on duckweed growth rate and also micronuclei occurrence. The effect on duckweed metabolism may become an important tool for environmental impacts evaluation caused by NP CuO. 11 ÉVALUATION DE LA TOXICITÉ DES NANOPARTICULES D OXYDE DE CHROME III CHEZ LES ALGUES CHLAMYDOMONAS REINHARDTII ET MICROCRUSTACÉS DAPHNIA MAGNA Da Costa 1, C. H., F. Perreault 2, S. Nanoparticule d oxyde de chrome III (NP Cr 2 O 3 ) est appliquée principalement dans l'industrie comme un pigment et un matériel réfractaire, mais son effet toxique sur les organismes vivants est peu connu. Ainsi, l'objectif de cette étude est d'évaluer l'effet de la NP Cr 2 O 3 chez les algues unicellulaires Chlamydomonas reinhardtii, chez la bactérie marine fluorescente Vibrio fischeri et chez le microcrustacé Daphnia magna. Les algues ont été exposées à des concentrations de 0,01; 0,1; 1 et 10 mg ml -1 de NP Cr 2 O 3, pendant une période de 72 heures. Il y a eu une évaluation de la production d espèces réactives d oxygène ROS (de l anglais Reactive Oxygen Species). Les tests avec la V. fischeri ont évaluées l inhibition de la luminosité de la bactérie marine pour une période d exposition de 30 minutes. Pour les tests avec la D. magna, ont été réalisés des essais de toxicité aigus et chroniques. Concernant l essai aigu (48 heures) les concentrations évaluées ont été de 0,01; 0,1; 0,5; 0,625; 1,25; 2,50; 5,0 et 10,0 mg ml -1. Concernant l essai chronique (21 jours), subséquent à l essai aigu, ont été testées les concentrations de 2,5; 5,0; 10,0; 25,0 et 50 mg L -1. Les résultats de ces tests chez les algues ont montré une augmentation significative au niveau de la ROS en 24 heures chez celles exposées aux NP Cr 2 O 3. Après 72 heures d exposition, le niveau de la ROS fut plus élevé que le contrôle dans une concentration de 10 mg ml -1. En rapport aux tests réalisés avec la V. fischeri, cette dernière s est montrée sensible à la NP Cr 2 O 3, présentant une CE50 30min moins que 0,08 mg ml -1. Le D. magna était sensible à la NP Cr 2 O 3, présentant une CE50 48h de 0,1 mg ml - 1. L essai chronique avec la D. magna a démontré l effet sur la reproduction et le développement à partir de la concentration de 10 mg ml -1, tandis que pour le paramètre de longévité, on n a seulement observé un effet que sur la plus grande dilution (50 mg ml -1 ). Avec les résultats obtenus, on a pu voir que la NP Cr 2 O 3 a présenté des effets toxiques aigus, chroniques et sur le métabolisme des organismes testés. Matias Laboratório de Toxicologia Ambiental, LABTOX, Departamento de Engenharia Sanitária e Ambiental, Universidade Federal de Santa Catarina, Campus Universitário, CEP: , Florianópolis, SC, Brazil Copper Oxide (CuO) has several applications, highlighting the incorporation as a biocide in the composition of antifouling paints for coating vessels and offshore platforms. The objective of this study was to evaluate and compare the aquatic toxicity of CuO in the form of nanoparticle (NP) and microparticle (MP), through acute and chronic toxicity test with microcrustaceans of freshwater Daphnia magna. Suspension tests were characterized by TEM, SEM, surface area, zeta potential and quantification of total dissolved Cu ions. The CuO NP (CE 50 = 22 mg L 1 ) had ten fold higher toxicity than CuO MP (CE 50 = mg L 1 ) for acute toxicity with D. magna and organisms exposed to this concentration were visualized by electron microscopy. Regarding chronic toxicity, both forms showed a statistically significant effect (p<0.05) for growth and reproduction parameters, morphological changes were observed such as no development of apical spine and malformation of the carapace of the exposed D. magna. Based on the characterization and the results shown it can be concluded that for the tests evaluated, the NP presented higher toxicity than the MP showing the need for comparative studies and toxicological evaluation for the correct classification once the CAS registry is the same for two forms of CuO. Elles sont utilisées par exemple comme agent de contraste (IRM, traçage) et pour délivrer des molécules thérapeutiques (nanovecteurs). Les réponses biologiques en présence des NPs résultent d interactions complexes qui dépendent de leurs propriétés physico-chimiques, du milieu biologique et du phénomène d adsorption de protéines qui forment une couche protéique appelée «corona». Il existe très peu d études qualitatives et quantitatives sur la formation et les conséquences de ce corona. Cependant, ces études sont nécessaires pour comprendre comment ces interactions peuvent être contrôlées afin que le potentiel des NPs pour les applications en nanomédecine puisse être pleinement exploité et utilisé de manière sécuritaire. Nous nous proposons, dans ce projet, de comprendre le mécanisme d adsorption des protéines à la surface des NPs du type oxyde de fer, afin d établir une corrélation entre leurs caractéristiques physicochimiques et leur biocompatibilité. Ainsi, plusieurs techniques de caractérisation de surface (DLS, Maldi TOF, FTIR ) et de mesure de l altération des fonctions cellulaires (par exemple, l activité mitochondriale et l intégrité de la membrane) ont été utilisées. Les réponses cellulaires aux NP d oxyde de fer ont été étudiées chez la lignée de cellules épithéliales alvéolaires humaines (A549). Nous avons démontré que la présence du corona réduit l'agglomération des nanoparticules. Nous avons également observé que la réponse cellulaire dépend des types des protéines adsorbées et surtout de leurs concentrations ou leurs affinités avec la nanoparticule concernée. Ces résultats montrent qu au-delà d'une caractérisation physico-chimique des NPs, une compréhension de l effet de la composition en protéines des milieux de culture ou des fluides biologiques (par exemple, le sang) sur le potentiel cytotoxique des NPs est cruciale pour l évaluation de la biosécurité des NPs développées pour les applications en nanomédecine. The exponential growth of nanotechnology and nanomaterials is particularly wondering environmental scientists. Current knowledge about effects of nanomaterials like silver nanoparticles (AgNPs) and carbon nanotubes (SWCN) on marine invertebrates is still largely insufficient and the field exposure levels are not well determined. Their physicochemical properties as well as their ability to cross biological membranes provides an interesting view on the way these contaminants can affect the early life of sea urchins. Some days after larvae metamorphosis the recently settled juveniles can optimize bioaccumulation by new internal transportation routes and their new feeding behaviour. Our experimental approach includes acute and chronic exposures of S. droebachiensis juveniles to four nominal concentrations of AgNPs and SWCN in order to investigate the effects of these nanomaterials and dissolved silver as a function of time. After 12h of acute exposition, individuals exhibit either oedema or severe necrosis over the body or their spines. During chronic exposition to AgNPs, tumor-like cell agglomerations appear into oral and sub-oral spines at 2 th week and necrosis thereafter (at 3 rd week). Confocal laser microscopy is used to assess abnormalities fine structures and stress oxidative at 96h. Values obtained from mortality and morphological analyses are expressed toward toxicity assessment criteria. Results provide new information about the uptake and effects of AgNPs, SWNC and ionic silver by echinoid juveniles as biological models in experimental conditions close to real coastal habitat. 14

15 STUDY OF PRO-INFLAMMATORY PROPERTIES OF DIFFERENT NANOPARTICLES USING IN VITRO AND IN VIVO MODELS De Liz, R., L. B. A. Leonardi, D. M. Gonçalves, D. Girard. Laboratoire de Recherche en Inflammation et Physiologie des Granulocytes, INRS-Institut Armand-Frappier, Université du Québec, Laval, Québec, Canada H7V 1B7 The use of nanomaterials and/or nanoparticles (NPs) has been largely increased in the last few years. Thus, several studies have been developed in order to validate the use of these different nanomaterials in various fields, including biomedical applications, such as monitoring and diagnostic technology, bioimaging, and drug delivery. However, in the past few years, some studies have demonstrated that different NPs induced pulmonary inflammation, as shown by investigating in vitro activation of pulmonary cells, as well as a variety of in vivo models in which neutrophils seemed to be the predominant leukocyte cells in lungs and in bronchoalveolar lavages. Neutrophils are the most abundant leukocytes in the peripheral blood. They play a pivotal role in the inflammation, since they are the first cells to arrive to the inflammation site. The neutrophils are the first line of innate immune defense against microbes by promoting phagocytosis and/or releasing toxic compounds, such as: reactive oxygen species (ROS), antimicrobial peptides, and proteolyitic enzymes from their granules. On the other hand, in physiological conditions, neutrophils have a short lifespan, since they undergo apoptosis in 12 hours. However, during the inflammatory process, the neutrophils are activated, and so they can stay longer in the inflammation site, resulting in persistent inflammation. Despite the fact that several studies have reported that different NPs induce neutrophil recruitment, the direct activation of these cells by a given nanoparticle (NP) is poorly understood. The aim of this study was to investigate the pro-inflammatory effects of different NPs, such as titanium dioxide (TiO 2 ), fullerenol (C 60 -OH), gold (Au), and silver (Ag) nanoparticles using in vivo and in vitro models. At non-cytotoxic concentrations (< 500 µg/ml) TiO 2 NPs showed to have pro-inflammatory properties, since they were effective in activating human neutrophils in vitro and inducing neutrophil migration in vivo. Also, C 60 -OH NPs were effective in potentiate the pro-inflammatory effect induced by lipopolysaccharide (LPS) in vivo. Moreover, Au NPs induced the apoptosis of human neutrophils in vitro, but they did not induce neutrophil recruitment in vivo. Finally, Ag NPs seem to have important immunomodulatory properties in human neutrophils, since they were effective in rapidly inducing an atypical cell death in a process that seems to be at least partially mediated by different caspases. 15

L utilisation des ressources forestières pour la production de nanomatériaux et notamment les nanotubes de carbone mono-parois (SWNT) a été proposée. Ces derniers présentent les caractéristiques les plus remarquables parmi les matériaux carbonés et sont étudiés de près pour un nombre de plus en plus important d applications notamment en remédiation et pour la vectorisation de médicaments. De plus, l ajout de groupements fonctionnels aux SWNT ou leur association à des polymères (fonctionnalisation) se développe de plus en plus car cela leur permet d être soluble dans l eau et facilite leur incorporation dans divers matériaux. La probabilité de retrouver ces différentes formes de SWNT dans l environnement, y compris le milieu aquatique, est donc vouée à augmenter alors qu il existe peu de données sur leur potentiel écotoxique notamment chez les invertébrés aquatiques. Par ailleurs, le domaine des nanotechnologies est en pleine expansion et certaines études ont déjà révélé les effets toxiques de certains NM en raison de leurs caractéristiques physiques et chimiques particulières. Le but de notre étude est d identifier à l aide de tests de toxicité aiguë et chronique les effets biologiques de SWNT bruts, purifiés ou fonctionnalisés sur la survie et la croissance d un amphipode d eau douce Hyalella azteca qui évolue dans les sédiments. Les nanomatériaux ayant tendance à sédimenter du fait qu ils soient peu solubles dans l eau, l utilisation d un organisme benthique comme modèle d étude semble pertinent. Les résultats préliminaires d exposition aux SWNT bruts et purifiés seront présentés. 16

17 IS SINGLE PARTICLE-ICP-MS THE METROLOGY TOOL FOR ENGINEERED NANOPARTICLES IN ENVIRONMENTAL MATRICES? Hineman, A., C. These materials are subsequently being released into the environment. Methods for detecting, quantifying, and characterizing these materials in complex matrices are critical for the eventual understanding of their implications to environmental quality and human health. This work describes Single Particle Inductively Coupled Plasma Mass Spectrometry (SP-ICP-MS) as a metrology tool in assessing the fate of engineered Nanoparticles in complex aqueous samples. Two case studies will be discussed; 1) Silver nanoparticle transformation in surface water, the effects of salinity, particle concentration, particle size, and particle surface chemistry. 2) Nanoscale Zero Valent Iron (NZVI) in environmental matrices employing the use of Dynamic Reaction Cell with Ammonia in order to reduce spectral background levels improving particle size detection. Single Particle (SP-ICP-MS) allows for the differentiation between ionic and particulate signals, quantitation of the ionic and particulate fractions, measurement of particle concentration (part/ml), estimation of particle sizes, identifies agglomeration, and creates a size distribution profile. 17

18 CARACTÉRISATION DES NANOPARTICULES PAR L ULTRACENTRIFUGEUSE ANALYTIQUE Diaz, L., K. J. D où l importance d évaluer le devenir, la mobilité et la biodisponibilité des NM une fois dans l environnement. La caractérisation et la quantification des NM dans l environnement sont des défis analytiques complexes étant donné leur dynamisme naturel (petite taille, grande réactivité et instabilité). L ultracentrifugation analytique (AUC) est une technique qui, à l origine, a été développée pour la caractérisation des protéines et d'autres macromolécules biologiques en solution. Néanmoins, il a été possible d étendre son utilisation à des NM métalliques (e.g. nag, quantum dots), des oxydes métalliques (e.g. ntio2, nzno) ou des nanoparticules polymériques. Dans cette étude, l'auc a été optimisée 1) pour la détermination des tailles de différents NM métalliques, 2) pour la caractérisation de petites nanoparticules polymériques (allosphères), commercialement utilisées pour favoriser la distribution des pesticides dans les champs agricoles. Le comportement des allosphères a été étudié en fonction du ph (6 à 8) à différente force ionique (10, 50, 100 et 500 mm NaCl et CaCl 2 ). Les résultats préliminaires, utilisant à la fois un détecteur d interférence et de fluorescence, indique que ces allosphères ont une taille d'environ 3.5 nm. Une faible variation de la taille a été observée en fonction de la force ionique et du ph ce qui suggère une certaine stabilité des allosphères. Le rôle de la matière organique naturelle sur l'agglomération de ces particules est actuellement en cours. 18

19 MILD AND SELECTIVE DEOXYGENATION OF GRAPHENE OXIDE USING LEWIS ACIDS IN SOLUTION Brisebois, P. P., R. Izquierdo, M. Siaj* Université du Québec à Montréal, Département de Chimie, C.P. 8888, Succ. Centre-ville, Montréal, QC, Canada (H3C 3P8) siaj.mohamed@uqam.ca Graphene oxide (GO) is a key precursor in the chemical functionalisation of graphene, a flat sheet molecule containing only sp2 carbon atoms having extremely high electrical and mechanical properties. It is a compound with no precise molecular weight having the averaged molecular formula C2.2H0.80O1.00, bearing alcohol and epoxide functions on the surface, and carbonyl and carboxylic acid functions mainly on the edge. The presence of oxygenated functions in the carbon matrix makes GO highly hydrophilic and dispersible in water. As a consequence, GO is suited for environmental and biological applications because of its low toxicity and high biocompatilbility. GO has proven to be useful for potential applications in the field of environment ranging from greenhouse gas adsorption, DNA sensing, bio culturing, toxicant adsorption, photocatalysis and capacitive desalination. Therefore in this work, we developed a novel process using a modified version of Hummer's method to prepare and exfoliate high quality GO in high yield from natural graphite. The process allows the modulation of the polarity of GO by selectively tuning its degree of oxidation and the size of the sheets in situ during the oxidation of graphite using chemosonication. Also, we investigated the mechanism of incorporation of the metallic ions in the matrix of GO using Lewis acids in solution. The results obtained from Fourier transformed infrared and 13C solid-state Nuclear Magnetic Resonance spectroscopies show that the Lewis acids used in this study are chemisorbed on GO and have preferential affinity for the carboxylic acid functions present on the edge of the sheets. In the presence of Lewis acids, GO undergoes a mild and selective decarboxylation and this strategy was employed to incorporate metallic ions derived from aluminium, copper, iron, magnesium, nickel, silver, tin and zinc. The nature of the Lewis acid influences the degree of deoxygenation of GO. This strategy was used to tune the degree of oxidation of the graphene oxide materials and to incorporate metallic ions in the matrix by controlling the parameters of the reaction (time, temperature, concentration, solvent). Briefly, we developed a process to produce GO-based materials with different physical properties and affinity for metallic ions. Our results showed that GO have a great affinity for metallic ions in solution and that GO undergoes a mild and selective decarboxylation in the presence of the Lewis acids. Finally, our work contributes to develop fundamental knowledge on the mechanism of action between GO and metallic ions in solution, hence allowing the development of devices for effective radionuclide and heavy metal removal, water remediation and pollution management. 19

20 ANALYSE DU PLOMB PAR PAPIER CARBONE FONCTIONNALISÉ Benjamin, O. D., D. Une méthode d analyse précise et peu coûteuse est très souhaitable pour déterminer la teneur en plomb de divers spécimens. Ces dernières années, une attention particulière a été portée au développement de nouveaux capteurs électrochimiques pour quantifier les ions des métaux toxiques au niveau de trace. Le choix d'un matériau d électrode et sa fonctionnalisation sont cruciaux pour le processus d'analyse. Le but principal de ce projet de recherche est de fonctionnaliser une électrode de papier carbone pour la détermination effective du plomb dans une solution aqueuse. À cet effet, le papier carbone activé, a été modifié avec de l'acide benzylphosphonique à partir de la réduction électrochimique des cations diazonium générés in-situ. La présence de la couche greffée a été prouvée et caractérisée par des techniques physico-chimiques telles que la spectroscopie du photoélectron-x et par des mesures électrochimiques. Les groupements benzylphosphoniques, liés de manière covalente à la surface de l électrode forment des complexes avec les ions Pb 2+ en solution. La quantité de plomb complexée est déterminée par l intégration de l aire sous le pic d oxydation ou de réduction d un voltammogramme cyclique. Ces matériaux fonctionnels présentent une limite de détection de 0,034 μg L -1 (S/N = 3) pour le plomb en solution. La détermination électrochimique du plomb a été examinée en variant différents paramètres : le temps de complexation, le ph de la solution aqueuse et la vitesse de balayage. The presence of heavy metals or emergent pollutants in wastewater or in watersheds exposes the population to chronic or acute accumulation of these contaminants affecting the population health with high economic cost. In this regard, this work aims the application of the nanoscience and nanotechnology tools in order to contribute with water/wastewater remediation contaminated with traditional pollutants. Therefore, we studied the remediation of metals ions, such as Pb 2+, Cd 2+, Cu 2+, Co 2+, Ni 2+ through the filtration of simulated ions effluents with fibrous cerium phosphate (CeP) and also its inedit nanocomposites obtained by interaction between CeP and functionalized multi-walled carbon nanotubes (MWCNT-Func). Herein, MWCNT-Func were functionalized by oxidation, thiolation and amidation reactions with 2- amino-2-hydroxymethyl-propane-1,3-diol (TRIS). In order to address the bio-fouling problem, we have developed a novel surface coating via polyelectrolyte layer-by-layer (LBL) selfassembly, functionalized with biocidal nanoparticles and antifouling/fouling release polymer brushes. The novel surface coating is prepared with polyelectrolyte [PEI: poly(ethylene imine); PAA: poly(acrylic acid) (PEI/PAA)] LBL films containing PEI coated silver nanoparticles (Ag- PEI), grafted either with hydrophilic [poly(sulfobetaine)] or with low surface energy (PDMS) polymer brushes. The presence of the silver nanoparticles at the functionalized surfaces is confirmed through surface analysis by X-ray photoelectron spectroscopy (XPS). Coating with both LBL films and sulfobetaine polymer brushes significantly increases the hydrophilicity of the membrane surfaces. Overall, surface modifications resulted in significant reduction of bacterial cell adhesion. In static bacterial adhesion tests (no pressure, no flow) with E. coli K12 MG1655, a normalized cell adhesion in the range of only 4 to16% on the modified membrane surfaces was observed. Modified surfaces containing silver nanoparticles were also noted to have strong antimicrobial activity. The membranes containing bilayers of PAA and 100% Ag-PEI were observed to achieve upto 95% inactivation of bacteria attached to the surface within 1 hour of contact time. Both antifouling and antimicrobial results suggest the potential of using this novel surface coating in controlling the RO membrane fouling. 23 POTENTIAL HEALTH EFFECTS OF WORKPLACE EXPOSURE TO ENGINEERED NANOPARTICLES, THE ROLE OF NANOEPIDEMIOLOGY El-Zein, M., D. Girard, M.-C. Rousseau INRS-Institut Armand-Frappier, Université du Québec, Laval, Québec, Canada The introduction of a novel class of materials, known as engineered nanoparticles (NPs), instigated new toxicological challenges raising the possibility of significant health side effects and safety concerns. Although there is acceptable experimental evidence in support of an association between exposure to NPs and adverse health effects, the question of whether human risks are actually involved has not been epidemiologically addressed. There is currently a great national and international interest in conducting nanoepidemiologic studies (epidemiological studies among nanotechnology workers) to investigate the effects of NPs on humans. In this presentation, we will provide a brief qualitative review of the current literature in terms of the main types, toxicity and exposure potential of NPs, postulated human health effects, and challenges to the conduct of nanoepidemiologic studies. The most critical limiting factor to the conduct of such studies has been the absence of concrete knowledge on which specific health outcomes to consider, though some have been speculatively inferred from findings in toxicological and epidemiologic studies of particulate air pollution and nanomaterials such as pulmonary and systemic inflammation, occurrence and worsening of chronic respiratory illness, pulmonary fibrosis, and lung cancer. As a first line of evidence gathering on expected health risks associated with exposure to work-related NPs, we intend to consider potential biological markers of effect as surrogate health endpoints which are indicative or predictive of several diseases. For the time being, this will provide us with fast insights on possible health effects of exposure to NPs (early inflammatory response) and raise interesting hypotheses accordingly (anticipated long term health effects). Results from this approach could further suggest and justify the conduct of more rigorous epidemiological designs. 23

24 ÉTUDE DE LA BIOSÉCURITÉ DES NANOTUBES DE CARBONE SIMPLES PAROI DOPÉS OU NON À L AZOTE Tabet, L., J.-P. Joarlette, Z. Ali Abbas, M. Welman, K. Maghni Centre de recherche, Hôpital du Sacré-Cœur de Montréal, 5400 Boul Gouin Ouest, Montréal, Québec, Canada, H4J 1C5 Introduction : Les nanotubes de carbone (NTC) sont des nanoparticules (diamètre < 100 nm) dont les propriétés physicochimiques favorisent leur utilisation dans plusieurs domaines technologiques. Cependant, les risques potentiels des NTC sur la santé humaine doivent être déterminés. L objectif de cette étude réalisée en collaboration avec la compagnie Raymor Nanotech (Québec) était d évaluer la biosécurité de leurs NTC. Méthodologie : Les cellules épithéliales alvéolaires humaines A549 ont été exposées à deux types de NTC: non dopés (RN-000) ou dopés à l azote (RN-003) produits par la compagnie Raymor Nanotech (Québec). La nanotoxicité a été déterminée par la mesure de la viabilité cellulaire, la mesure de l apoptose/nécrose par diverses techniques (e.g. cytométrie en flux) et l induction d un stress oxydant. Les cellules ont été incubées avec les NTC ( µg/ml) durant 24h. Les points quantiques (PQ) ont été utilisés comme témoin positif de nanotoxicité. Résultats : À l inverse des PQ, RN-000 et RN-003 n ont provoqué aucune altération de la viabilité cellulaire ni induit d apoptose/nécrose chez les cellules A549, même à une concentration de 1000 µg/ml. Malgré l absence de nanotoxicité, ces NTC ont induit des changements morphologiques des cellules A549, et un stress oxydant qui a été contrebalancé par les cellules. Fait intéressant, cette interaction cellules/ntc a induit une diminution de leur prolifération au cours du temps. Conclusion : L'absence de toxicité du RN-000 et RN-003 chez les cellules A549 indique une certaine biosécurité de ces NTC. Cependant, ils interagissent avec les cellules en provoquant un ralentissement de leur prolifération ce qui pourrait se traduire in vivo par une altération de la régénération de l épithélium alvéolaire, le cas échéant. Ainsi, l extrapolation de ces études expérimentales à l être humain indiquerait l existence d un risque potentiel à la santé lors de l exposition pulmonaire. Cependant, il est complètement incertain si l exposition aux nanoparticules manufacturées provoquera des problèmes respiratoires transitoires ou donnera naissance à un nouveau type de maladie respiratoire. Cette problématique se complique dans le cas de personnes atteintes d une maladie respiratoire, telle que l asthme. Cette présentation sera principalement axée sur les connaissances actuelles et les futures directions dans la recherche expérimentale sur l influence de l exposition pulmonaires aux nanoparticules manufacturées sur le développement et l exacerbation de l asthme. Contact lhocine.yahia@polymtl.ca Recent advances in nanotechnology have led to the development of magnetic nanoparticles (NMs) iron oxide, including magnetite (Fe 3 O 4 ), for drug delivery, magnetic resonance imaging (MRI), repair tissue, hyperthermia and cell separation. Biomedical applications such characteristics are due to the MNs, such as a narrow size distribution, a superparamagnetism, a magnetic moment and a high surface to volume ratio. Many synthetic methods have been used to produce Fe 3 O 4 nanoparticles, such as co-precipitation, high temperature decomposition, microemulsions, etc. However, the particles thus obtained are often non-uniform size, incomplete crystallinity inhomogeneous composition and aggregate easily due to the magnetic attraction. Recently, an interesting alternative to these synthetic nanomaterials, called magnetosomes, was found in magnetotactic bacteria. The synthesis of magnetosomes by a biological process promises benefits in terms of controlling the crystal size and morphology, and superparamagnetic behavior at room temperature. Our project aims at delivering nitric oxide (NO) anti-biofilm via NMs to prevent nosocomial infections related to implants. In this work, in order to choose the appropriate NMs, we compared the morphological and magnetic physico-chemical properties of iron oxide nanomaterials to those of magnetosomes extracted from the bacterial type strains Magnetococcus sp. MC-1. The techniques used for the characterization and comparison of their properties were: transmission electron microscopy (TEM), Fourier transform infrared (FTIR), vibrating sample magnetometer (VMS) spectroscopy and X-ray photoelectron (XPS). The interactions between dissolved organic matter (DOM) and AgNPs tend to influence and/or control the dissolution of AgNPs, which is an important environmental transformation affecting silver mobility, bioavailability and toxicity in natural waters. The aim of this work was to investigate how AgNPs react with DOM under environmentally relevant conditions. Particle size characterization was determined by Dynamic Light Scattering (DLS; Zetasizer Nano S, Malvern). Zeta potential measurements were performed to evaluate the stability of nanoparticles using the same instrument. Free Ag + ions dissolved in solution in the absence or presence of SRHA were determined by dialysis experiments, and compared with an ion-exchange technique (IET). The role of DOM in the stability and dissolution of AgNPs was evaluated initially under controlled laboratory conditions. Mesocosm experiments will also be conducted to monitor the fate of silver nanoparticles. Preliminary results suggest that Z- average size of poly(allylamine) coated silver nanoparticles, which have a nominal particle size of 15 nm (referred as AgNP-PAAm15), was observed to significantly increase with increasing concentrations of Suwannee River Humic Acid (SRHA). In addition, dissolved Ag concentration was observed to be more than 90% of total AgNP-PAAm15 concentration (1.2 mg/l) after 48 h incubation without SRHA in a simplified algal culture medium. 27
