2 Abstract The aim of this thesis was to explore the statistical physics and the mechanical properties of individual biomolecules, to study their interactions, and to develop new techniques based on atomic force microscopy (AFM). First, we exploit the high resolution imaging of AFM over long DNA molecules adsorbed on a surface to measure the average end-to-end distance as a function of the DNA length, along with its full distribution function. The latter is almost never determined experimentally, and is not available with ensemble techniques, such as light scattering. We found that the scaling exponents are closer to the values predicted by the polymer theory for a 3D self-avoiding random walk (SAW) than for a 2D SAW. These results suggest that the adsorption process of the DNA molecules is akin to a geometric projection from 3D to 2D, known to preserve the scaling properties of fractal objects of dimension d f < 2. Second, taking advantage of the possibility of AFM to also manipulate biomolecules, we investigate the specific interactions between the glycoprotein avidin and the vitamin biotin, the ribonuclease barnase and its inhibitor barstar, and the West Nile Virus surface glycoprotein E and the laminin binding protein. The dependence of the most probable unbinding force on the force loading rate has revealed details of the energy landscapes of the unbinding process of these highly efficient and specific interactions. Beside this standard method, we have measured the lifetime of the avidin-biotin bond, kept stretched at a constant force, as a function of this force, and combined this approach with a small dithering of the AFM tip in order to further explore the avidin-biotin energy landscape. Finally, the viscoelastic properties of biomolecules, such as dextran and titin, have been studied using standard force spectroscopy combined with a small oscillation of the AFM tip. A theoretical description for excitation frequencies near the first and the second resonance of the cantilever has been developed to be able to extract, from the amplitude and phase measurement of the oscillations, the stiffness and friction coefficients of the biomolecule being stretched. We conclude that an excitation frequency of 3 khz gives reliable results for the stiffness measurement whereas frequencies of khz are more appropriate for the dissipation measurements. 3 Résumé Le but de cette thèse était d explorer les propriétés statistiques, mécaniques, et d interactions des biomolécules, en dévelopant de nouvelles techniques basées sur la microscopie à force atomique (AFM). Nous avons exploité la très haute résolution de l imagerie par AFM sur de longues molécules d ADN adsorbées sur une surface pour mesurer la distance bout-à-bout moyenne en fonction de la longueur de l ADN, ainsi que sa fonction de distribution. Cette dernière est rarement déterminée expérimentalement, car impossible à déterminer avec des techniques traditionnelles d ensemble telle que la diffraction de la lumière. Nous avons trouvé que les exposants d échelle sont plus proches des valeurs prédites par la théorie des polymères en 3D que en 2D. Ces résultats suggèrent que notre processus d adsorption peut être apparenté à une projection de 3D sur 2D, connue pour préserver les propriétés d échelle pour des objets fractals de dimension d f < 2. Profitant également de la possibilité offerte par l AFM de manipuler des biomolécules, nous avons étudié les interactions spécifiques agissant entre la glycoprotéine avidine et la vitamine biotine, le ribonucléase barnase et son inhibiteur barstar, la glycoprotéine E du virus du Nil occidental et de la protéine laminine. La dépendence de la force de rupture la plus probable sur la vitesse d étirement a révélé des détails sur le paysage énergétique des processus de rupture de ces interactions très efficaces et spécifiques. A côté de cette méthode standard, nous avons également mesuré le temps de vie des liaisons avidinebiotine en fonction de la force appliquée sur celles-ci et combiné cette approche avec une petite oscillation du levier AFM. Finalement, les propriétés viscoélastiques de biomolécules, tels que le dextran et la protéine titine, ont été étudiées en utilisant la spectroscopie de force conventionnelle combinée à une petite oscillation du levier AFM. Une description théorique pour des fréquences d excitation proche de la première et deuxième résonance du levier a été développée pour pouvoir extraire, à partir des mesures d amplitude et de phase des oscillations, la rigidité et la friction de la biomolécule étant étirée. Nous avons conclu qu une fréquence d excitation de 3 khz donne des résultats fiables pour les mesures de la rigidité tandis que des fréquences de khz sont plus appropriées pour les mesures de friction.
