Le Laboratoire de Biochimie et de Génie Enzymatique des Lipases de l ENIS est spécialisé dans l étude des relations structure-fonctions des enzymes impliqués dans la dégradation des lipides et responsables de l obésité. Le laboratoire s intéresse à l étude des enzymes de différentes et à l étude de certaines applications biotechnologiques. Les travaux de recherche au laboratoire sont axés sur plusieurs disciplines : biochimie, génie génétique, biologie moléculaire, modélisation, microbiologie... Le laboratoire est constitué de deux équipes de recherche : 1- Enzymes lipolytiques de microorganismes et applications biotechnologiques. Les principales activités de recherche de cette équipe sont : - Isolement et identification de microorganismes à partir de biotopes extrêmes Tunisiens et/ou marocain et optimisation des conditions de production des lipases. - Purification et caractérisation biochimique et moléculaire des enzymes isolées. - Modélisation et/ou cristallisation des lipases. - Amélioration de la biodégradabilité des effluents d industrie agroalimentaires riches en lipides (industrie laitière, transformation de la viande et des produits de la mer) par addition d enzymes lipolytiques. - Recherche d inhibiteurs de lipase qui peut être utilisé comme un médicament anti-obésité. - Valorisation des sous produits agricoles et d industries alimentaires (grignon d olive, son de blé, écorce d orange, écorce de sésame.) comme support et /ou substrat pour la production d enzymes lipolytiques par fermentation en milieu solide. - Collaboration avec des industriels pour tester l incorporation des molécules à haute valeur ajoutée présentant le label «bio». 2- Enzymes lipolytiques d animaux inférieurs terrestres et marins Les principales activités de recherche de cette équipe sont : - Caractériser les enzymes digestifs d animaux terrestres inférieurs (scorpion ) et produire des anticorps spécifiques pour la cytolocalisation de ces enzymes. - Explorer les systèmes digestifs des animaux marins pour pouvoir dégager des traits communs entre les systèmes lipolytiques des différentes classes d animaux marins (poissons osseux, poissons cartilagineux, arthropodes ). - Isoler les gênes codant pour les lipases de scorpion, de raie et de l anguille pour déterminer les séquences complètes en acides aminés de ces enzymes digestives et établir les relations phylogénétiques avec les autres familles de lipases. - Explorer les possibilités d utilisation un de ces enzymes lipolytiques d animaux inférieurs comme agents thérapeutiques potentiels.
