2 Abstract ABSTRACT Enzymes are the chemical machinery of nature. These protein catalysts promote diverse chemical reactions with unbeatable rates and selectivities 1. As a consequence, they are already a center of attention in the chemical and pharmaceutical industries, where viable alternatives to expensive, polluting and noncrenewable catalysts are continually sought. The possibility of engineering, redesigningormodelingenzymesfromscratchisanimportantasset. To exploit the advantages of biocatalysts from nature and metalccatalyzed reactionsdevelopedinthelaboratory,thefieldofmetalloenzymeshasemerged.it aimstocombinethebroadsubstratescopeofhomogeneouscatalysiswiththelarge turnover numbers, mild reaction conditions, and high evolvability of enzymes. In thisthesisweexploredthefeasibilityofexploitinggold(i)forthispurpose.overthe last decades many studies have demonstrated that gold(i) complexes with triphenylphosphine and NCheterocyclic carbenes are robust catalysts for diverse transformations. Several different strategies for incorporating a metal ion into a protein scaffold are conceivable. One can replace the metal cofactor of a natural metalloc enzymeorcovalentlybindametalioncomplextoaproteinscaffoldthatdoesnot normally harbor a metal. We focused on the soccalled supramolecular strategy, whichtakesadvantageoftheaffinityofenzymesfornoncproteinogenicmolecules thatcanserveasligandsforthemetalcenter. ix

3 Abstract InChapter2wetriedtoexploitanaturallyoccurringcofactortocoordinate gold(i) and anchor it at an enzyme active site. The vitamin thiamin, a cofactor utilized by a large family of enzymes, is similar in structure and reactivity to NC heterocycliccarbenes.thefirstsynthesisofamonogold(i)ccarbenewithathiamin analogue is described. The reaction can be carried out with a benzyl thiazolium derivativeinasinglestepunderaqueousconditions.xcraycrystallographicanalysis confirmedthatthetarget structure was obtained.thisthiazoliumgold(i)carbene complexwasalsoshowntocatalyzethe5cendocdigcarbocyclizationofanacetylenic dicarbonylcompound.effortstobindthecatalysttotwomembersofthethiaminc dependentenzymefamilyaredetailed.however,despitegeneticengineeringofthe active site of transketolase and benzaldehyde lyase to accommodate the cofactor analogue,thebindingofthemetalccatalysttotheenzymeswastooweaktoafforda functionalsupramolecularcomplex. ToengineerametalCcomplexwithhigheraffinityforaproteinhost,wetook advantageoftheevercgrowingnumberofenzymeinhibitors.inchapter3,aclassof molecules that inhibits chymase, a human protease involved in a variety of inflammation processes, was used as a ligand for gold. The imidazolone core of a published nanomolar inhibitor was replaced with an imidazolium salt. Deprotonation of the latter, followed by formation of the bis silverccarbene and addition of dimethylsulfide gold(i) chloride gave a mono gold(i)ccarbene complex by transmetallation. Precursors of the carbene, in which a carboxylate side chain important for chymase recognition was protected as an ester, were shown to be active as catalysts in the hydroalkoxylation of an allene. The deprotected gold(i)c carbenederivativealsobindschymasewithasimilaric50(6μm)comparedtothe templateinhibitor,thusdemonstratingthefeasibilityofaddingagold(i)atomtothe inhibitorwithoutdeleteriouseffectsonenzymebinding.unfortunately,thisgold(i)c carbene was not active, likely because the carboxylic acid group in the organic ligand forms a salt bridge with the cationic gold, inactivating the catalyst. 4 Abstract hypothesized that binding of the complex to chymase might unleash its catalytic potentialsincethecarboxylicacidinquestionshouldengageinastronginteraction withtwolysineresiduesattheactivesite.becausewewerelimitedbytheamounts of expensive recombinant chymase available, however, only a 200Cfold lower concentration of the proteincmetalccomplex compared to the reaction without enzymecouldbeachieved.undertheseconditions,noactivitywasdetected.our results demonstrate that the large pool of existing enzyme inhibitors are a potentiallyinterestingsourceofligandstoanchormetalionsatenzymeactivesites. Nevertheless, practical implementation of this idea will require proteins that are easy to engineer and available in large quantities at reasonable prices. In the present case, methods for improving the recombinant production of human chymasewillbeneededtorealizetheoriginalgoals. Guidedbytheresultsofthetwopreviouschapters,weturnedtothebiotinC streptavidinsysteminchapter4.thebindingofbiotintostreptavidinisthetightest nonccovalent interaction known to date, and both entities are easily accessible. Whitesides made the first artificial rhodium metalloenzyme with this system and Warddescribedhowinteractionsbetweenresiduesoftheproteinandtheappended metalcomplexcanenhanceefficiencies.ourexpertiseinmakingthiazoliumgold(i)c carbenes in aqueous media was exploited to prepare a biotinylated benzylthiazolium derivative that served as a precursor for making a gold(i)c carbene. The thiazolium derivative that was chosen has an aldehyde at the C5 position,whichreactsdirectlywithbiotinhydrazide.althoughthiscomplexhasnot yet been fully characterized, and probably exists as a mixture of mono and bis gold(i)ccarbenespecies,itwascatalyticallyactiveinthepresenceofthestreptavidin host. The hydroalkoxylation of an allene under aqueous conditions proceeded slowlyin9%yieldand9%enantiomericexcess.thelowamountofmonogold(i)c carbeneinthemixturemayaccountforthelowconversion.furthermore,because theiminebondhastworotamericforms,themetalionprobablyoccupiesdifferent xi

5 Abstract locations on the protein, with different residue environments that may confer oppositeselectivities.althoughmuchworkremainstobedone,theseexperiments provide the first example of a goldcdependent enzyme, demonstrating that gold catalysis is compatible with a protein scaffold. We are confident that it will be possible to achieve higher yields and selectivities with asingle species of a mono gold(i)ccarbene. Inthisthesis,wedevelopedanewoneCsteproutetomonogold(I)Ccarbenes from water soluble thiazolium salts. Such complexes may lead to interesting findings in homogeneous and enzymatic goldccatalysis. We also showed that gold catalystsarebiocompatible.theirabilitytoexpandthescopeofnaturalenzymatic reactionshasnotyetbeenrealized,butproteinscaffoldsmaysomedaycomplement smallchiralligandsandcounterionsasameanstoconferhighratesandselectivities ongoldccatalyzedreactions. xii

Lathiamine,unemoléculesemblableauxcarbènesNChétérocycliquestantpar sa structure que par sa réactivité, est le cofacteur utilisé par toute une famille d enzymes. Le Chapitre 2 décrit comment un ion d or(i) à été coordonné à ce cofacteur naturel afin de l ancrer dans le site actif d une enzyme. La synthèse du monoccarbèneàl or(i)d unanalogueàlathiaminefuteffectuéeenuneseuleétape en conditions aqueuses. Une analyse cristallographique aux rayons X a confirmé l obtentionducomplexesouhaitéetnousavonsdémontréquececatalyseuràl or(i) accélèrela5cendocdigcarbocyclisationd uncomposédicarbonylacetylenique.afin de remodeler les sites actifs de deux enzymes thiaminecdépendantes (la transkétolase et benzaldehyde lyase) pour y accommoder le nouveau cofacteur, nous avons eu recours à l ingénierie génétique, ce qui n a malheureusement pas résulté en une affinité suffisante pour conférer au système des propriétés supramoléculairesfonctionnelles. Le Chapitre 3 détaille la façon dont des inhibiteurs enzymatiques ont été exploitéspourcontournerlesproblèmesd affinitérencontrésprécédemmententre l enzyme et le métalloccomplexe. La chymase est une protéase humaine qui participe à de nombreux processus inflammatoires; elle est inhibée à des concentrationsnanocmolairesparuneclassed inhibiteursspécifiquesnoncnaturels baséssuruneimidazolone.nousavonsremplacécenoyauparunseld imidazole quiaprèsdéprotonation,formationdubisccarbèned argentetadditiondesulfure de diméthyl d or(i) a produit le monoccarbène à l or(i) par transmétallation. Des précurseurs du carbène, protégés par un acide carboxylique ont démontré une activité catalytique pour la réaction d hydroalkylation d un allène. Dans ces conditions, aucune activité ne fût détectée.nosrésultatsdémontrentquelapanoplied inhibiteursenzymatiquesest une source intéressante de potentiels ligands servant à ancrer un ion métallique danslesiteactifd uneenzyme.néanmoins,pourmettrecetteidéeenpratique,des enzymes faciles à modifier et disponibles en grandes quantités à moindres coûts sontindispensables.dansnotrecas,ilseraitnécessaired optimiserlesméthodesde productionrecombinantedelachymasepouratteindrenotreobjectifinitial. Grâce aux leçons apprises dans les deux Chapitres précédents, nous nous sommes basés sur le système biotine/streptavidine dans le Chapitre 4. La liaison entrelabiotineetlastreptavidineestlaliaisonnonccovalentelaplusforteconnueà cejouretlesdeuxcomposantesdusystèmes obtiennentfacilementetàbasprix. C estenutilisantcesystèmequewhitesidesdéveloppalapremièremétalloenzyme artificielle au rhodium dans laquelle Ward démontra que les interactions entre résidusprotéiquesetcomplexemétalliquepouvaientrenforcerl activitécatalytique. Basé sur notre expertise à synthétiser des carbènes de thiazolium à l or(i) en conditions aqueuses, nous avons préparé un dérivé du benzylthiazolium biotinylé avecungroupealdéhydeenpositionc5pouvantréagiravecl hydrazidedebiotine, entantqueprécurseurducarbèneàl or(i).mêmesilecomplexeobtenun apasété caractérisédefaçonapprofondie(ilpourraits agird unmélangedemonocetbisc carbène), une activité catalytique en présence de streptavidin fut détectée. L hydroalkylationd unallèneenconditionsaqueusessedéroulalentementavecun xv

Il est possible que la faible quantitédemonoccarbèneàl or(i)soitresponsabledufaibletauxdeconversion.vu quelaformationdelaliaisoniminegénèredeuxrotamères,l ionmétalliquepeutse trouver dans deux environnements différents au sein de la protéine pouvant conférer des sélectivités opposées. Malgré une activité catalytique noncoptimale, nous avons démontré dans ce chapitre que la catalyse à l or et les structures protéiquessontcompatibles. Danscettethèse,nousavonsdéveloppéunenouvellevoiesynthétiquepour lescarbènesàl or(i)enuneseuleétapeàpartirdeselsdethiazoliumsolublesdans l eau. Nous avons démontré que les catalyseurs à l or sont biocompatibles ce qui pourraitconduireàd intéressantsdéveloppementsencatalyseenzymatiqueàl or. Nous espérons que dans un avenir proche les structures protéiques puissent être utilisées en complément des ligands organiques et contrescions chiraux comme moyend augmenterlestauxdeconversionetconférerdessélectivitésélevéesaux catalyseursàl or. xvi
