2 Abstract Real-time monitoring of neurotransmitters is of utmost importance for understanding the functioning of the brain. Especially L-glutamate and choline play a major role in chemical signalling and are highly involved in cognitive functions such as learning, memory and behaviour. In this thesis, silicon microprobe arrays were realised comprising several microelectrodes with a size of µm 2. The microtechnological fabrication process allows an application-tailored design to position the electrodes at defined target regions in the brain. Microprobes with a length up to 8 mm and a cross-section of µm 2 have been fabricated allowing minimally invasive implantation at low tissue damage to be achieved. L-glutamate and choline are detected by the amperometric detection of peroxide using the microelectrodes coated with an enzymatic membrane. An array-compatible method for spatially controlled and parallel membrane deposition by electrochemically aided adsorption and chemical co-cross-linking with glutaraldehyde was developed. The enzymatic membrane deposition is followed by electropolymerisation of m-phenylenediamine to deposit a semipermeable membrane rejecting electroactive interferents such as ascorbic acid and dopamine that are endogenously present in the extracellular fluid. The resulting biosensors show adequate characteristics in sensitivity, detection limit as well as functional and storage lifetimes for acute monitoring of the neurotransmitters in brain tissue. The results of an extensive in vitro assessment and in vivo functional tests are presented. In array configuration, the biosensors allow simultaneous multi-site recordings of one analyte from different brain regions or multi-analyte recordings within defined spatial brain areas. Additionally, microchannels were integrated with the biosensor arrays to perform chemical stimulation by the local delivery of neuroactive substances: SU-8 microinjector arrays comprising several microfluidic channels or the direct integration of microchannels into the silicon shaft were realised. Both allow a precise positioning of the fluidic outlet relative to the biosensors and lowvolume injections. The biosensors integrated with the delivery channels enable the bilateral interaction with neuronal tissue on the neurochemical level at required temporal

3 and spatial resolution. This thesis is a part of the European integrated project NeuroProbes aiming at realising three-dimensional arrays of multi-functional microprobes, assembled in a modular way to perform in vivo electrophysiological and biological measurements. The biosensor is one of the functionalities to be integrated in this platform. The result is a new device for neuroscientists that will allow most complete 3D mapping of neuronal circuitry and relate these complex signalling mechanisms to observable behaviour. 4 Résumé La mesure en temps réelle de neurotransmetteurs est de la plus haute importance pour comprendre le fonctionnement du cerveau. Le L-glutamate et la choline joue un rôle majeur dans la transmission de signaux chimiques et sont fortement impliqués dans les fonctions cognitives telles que l apprentissage, la mémoire et le comportement. Dans cette thèse, une matrice de micropointes en silicium comprenant plusieurs microélectrodes de µm 2 fût réalisée. Leur conception sur mesure par des méthodes de microfabrication permet le placement des électrodes dans des régions cibles prédéfinies du cerveau. Des micropointes d une longueur maximale de 8 mm et d une section de µm 2 furent fabriquées afin de permettre leur implantation en minimisant les lésions provoquées aux tissus cérébraux. Le L-glutamate et la choline sont mesurés ampérométriquement par détection de peroxyde généré par une membrane enzymatique couvrant les microélectrodes. Une méthode de déposition de membranes par adsorption électrochimique assistée et co-réticulation avec du glutaraldehyde fût développée. Elle est compatible avec la matrice de pointes pour le dépôt localisé et en parallèle. Le dépôt de la membrane enzymatique est suivi par l électro-polymérisation de m- phenylenediamine pour obtenir la déposition d une membrane semi-perméable qui bloque des interférents électroactifs tels que l acide ascorbique et la dopamine, ces derniers étant présent dans le fluide extracellulaire. Les biocapteurs obtenus présentent des caractéristiques adéquates en termes de sensibilités, limite de détection ainsi que de durée de vie de conservation et en opération pour la détection précise des neurotransmetteurs dans les tissus du cerveau. Les résultats d une vaste évaluation in vitro et de tests fonctionnels in vivo sont présentés. En configuration en matrice, les biocapteurs permettent la mesure simultanée d un analyte dans plusieurs régions du cerveau ou de plusieurs analytes dans une zone précise. De plus, des microcanaux furent intégrés dans la matrice de biocapteurs permettant une stimulation chimique par la distribution locale de substance neuroactive: Des micro-injecteurs en SU-8 avec plusieurs canaux microfluidiques

5 ou l intégration directe des microcanaux dans les pointes en silicium furent réalisés. Les deux solutions permirent un positionnement précis de l extrémité du canal par rapport au biocapteur pour une injection de petits volumes. Les biocapteurs avec les microcanaux intégrés permettent une interaction bilatérale avec le tissu neuronal au niveau neurochimique à la résolution spatiale et temporelle requise. Cette thèse fait partie du projet européen NeuroProbes visant à la réalisation de matrices tridimensionnelles de micropointes multifonctionnelles assemblées de façon modulaire pour des mesures électrophysiologiques et biologiques in vivo. Le biocapteur est l une des fonctionnalités à être intégré dans cette platforme. Le résultat est un nouveau dispositif pour la neuroscience qui permettra une mesure tridimensionnelle fortement avancée des circuits neuronaux et de lier ces mécanismes complexes de transmission à des comportements observables.
