1 Dako Liquid Permanent Red for Immunohistochemistry and In Situ Hybridization English Code K0640 Intended use For In Vitro diagnostic use. Liquid Permanent Red (LPR) substrate-chromogen system is intended for use in immunohistochemical and in situ hybridization staining procedures utilizing alkaline phosphatase. LPR forms a permanent red reaction product at the site of the target antigen or nucleic acid, which can be visualized with standard optical light or fluorescent microscopy (using Texas Red or Rhodamine filters). 1 Reagents provided The following materials, sufficient for preparation of 30 ml or 110 ml of substrate-chromogen solution, are included: Quantity Description Small Volume 1 ml Liquid Permanent Red Chromogen 30 ml Liquid Permanent Red Substrate Buffer Large Volume 3 ml Liquid Permanent Red Chromogen 110 ml Liquid Permanent Red Substrate Buffer Precautions Storage Reagent preparation 1. For professional users. 2. Avoid ingestion, inhalation, and skin contact. If skin contact occurs, immediately flush affected area with water. 3. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin. 4. Unused solution should be disposed of according to local, State and Federal regulations. 5. Safety Data Sheet (SDS) available to professional users upon request. Dako Liquid Permanent Red should be stored at 2 8 C. If reagents are stored under any conditions other than those specified in the package insert, they must be verified by the user. Do not freeze. 1. Allow Substrate Buffer and Chromogen to equilibrate to room temperature before use. 2. Dispense 3 ml of LPR Substrate Buffer into a test tube. Add one drop LPR Chromogen to the tube. Mix well. 3. The prepared Substrate Buffer-Chromogen reagent should be used within 30 minutes. For best results, prepare immediately before use. 4. Larger volumes of Substrate Buffer-Chromogen solution may be prepared by mixing one drop of Chromogen in each 3 ml of Substrate Buffer required. ( ) P04032EFG_01_K0640/ p. 1/7

2 Procedure Following incubation with an alkaline phosphatase-labelled reagent, rinse off excess reagent with buffer from a wash bottle (do not focus stream directly on specimen area). Place specimen in a buffer bath for approximately 5 minutes. Following washing, tap off excess buffer and carefully wipe slide around specimen. STEP 1. Cover specimens with prepared LPR solution. Incubate 5 20 minutes. Optimal incubation time may vary with target concentration and/or specimen fixation and should therefore be determined by the individual laboratory. Rinse gently with reagent-quality water. STEP 2. Counterstain, if desired, with hematoxylin. (See below for recommendations.) Incubate for 2 5 minutes, depending on the strength of the hematoxylin used or the desired intensity of the counterstain. Recommendations: Dako Hematoxylin (code S3302) for 2 minutes is recommended for tissue specimens. Dako Hematoxylin (code S3309) for 4 minutes is recommended for cytology specimens. Dako Automation Hematoxylin (code S3301) is recommended for use with the Dako Autostainer. Rinse gently with reagent-quality water and then place in a water bath for 2 5 minutes. Ensure all residual hematoxylin has been cleared. STEP 3. Coverslip with permanent or aqueous mounting media. For aqueous mounting, mounting media such as Dako Glycergel Mounting Medium (code C0563), Faramount Aqueous Mounting Medium (code S3025) or Ultramount (code S1964) is recommended. For non-aqueous, permanent mounting, it is recommended that slides are air-dried completely at room temperature or air-dried for 1 hour at 60 C, and t hen incubated in xylene or a xylene substitute for 5 minutes. Note that in some instances small, punctate artifacts may be observed with slides subjected to air drying prior to application of coverslips. This can be addressed by treating the slides according to one of the following procedures: Option 1: Direct transfer of tissue slides from the Autostainer instrument to alcohol, which may be either: a) 99% alcohol with incubation for maximum 2 min, or b) Three successive 100% alcohol baths, 10 dips in each. Then slides should be dipped 10 times in each of two successive xylene baths followed by permanent mounting. Option 2: Direct transfer of tissue slides from the Autostainer instrument to a graded alcohol series, but only for 30 sec in each step and without xylene exposure, followed by permanent mounting. Longer alcohol and/or xylene exposures can lead to chromogen extraction and hence inferior staining. It is noted that these procedures have not been tested on every primary antibody, and should therefore only be interpreted as guidelines. Accordingly, the performance of the adjusted dehydration protocol followed by permanent mounting must be verified to ensure that it is still valid regarding the primary antibody in question. Each primary antibody in use should be evaluated with the adjusted dehydration protocol to confirm that the staining pattern is identical to the staining pattern described in Performance Characteristics of the primary antibody. Results Use of the Liquid Permanent Red Substrate Buffer-Chromogen yields a permanent red reaction product at the site of the target antigen or nucleic acid which can be visualized with standard optical light or fluorescent microscopy (using Texas Red or Rhodamine filters). 1 For proper interpretation, positive and negative controls should be run alongside unknown specimens. Positive controls serve as indicators that specimen processing and handling were carried out correctly. Negative controls are useful for assessing nonspecific staining. Endogenous alkaline phosphatase activity in some tissues may yield false-positive results. This endogenous activity may be inhibited by use of Dako Dual Endogenous Enzyme Block (code S2003) or Levamisole (code X3021). Levamisole does not inhibit the intestinal or placental form of alkaline phosphatase and may not be useful for staining of specimens containing these isoenzymes. La fiche de données de sécurité destinée aux utilisateurs professionnels est disponible sur demande. Le Liquid Permanent Red (Rouge permanent liquide) de Dako doit être conservé entre 2 et 8 C. Si les r éactifs sont conservés dans des conditions autres que celles indiquées dans la notice, celles-ci doivent être validées par l'utilisateur. Ne pas congeler. 1. Laisser le tampon de substrat et le chromogène atteindre la température ambiante avant utilisation. 2. Verser 3 ml de tampon de substrat LPR dans un tube à essai. Ajouter une goutte de chromogène LPR dans le tube. Bien mélanger. 3. Le réactif tampon de substrat chromogène ainsi préparé doit être utilisé dans les 30 minutes. Pour de meilleurs résultats, préparer immédiatement avant utilisation. 4. De plus grandes quantités de solution tampon de substrat chromogène peuvent être préparées en mélangeant une goutte de chromogène pour 3 ml de tampon de substrat, selon la quantité requise. 4 Procédure Après incubation avec un réactif marqué à la phosphatase alcaline, rincer l'excès de réactif avec le tampon contenu dans un flacon de rinçage (ne pas diriger le jet directement sur l'échantillon). Placer l'échantillon dans un bain de tampon pendant environ 5 minutes. Après le rinçage, tapoter pour éliminer l'excès de tampon et essuyer avec soin le contour de l'échantillon sur les lames. ÉTAPE 1 ÉTAPE 2 Couvrir les échantillons de solution LPR. Incuber pendant 5 à 20 minutes. Le temps d'incubation optimal peut varier en fonction de la concentration cible et/ou de la fixation des échantillons ; il doit donc être déterminé par chaque laboratoire. Rincer doucement à l'eau de qualité réactif. Contre-colorer, si nécessaire, à l'hématoxyline. (Voir ci-après les recommandations). Incuber pendant 2 à 5 minutes, en fonction de la puissance de l'hématoxyline utilisée ou de l'intensité de contre-coloration souhaitée. Recommandations : Il est recommandé d'utiliser le produit Hematoxylin de Dako (réf. S3302) pendant 2 minutes pour les échantillons tissulaires. Il est recommandé d'utiliser le produit Hematoxylin de Dako (réf. S3309) pendant 4 minutes pour les échantillons cytologiques. Il est recommandé d'utiliser le produit Automation Hematoxylin de Dako (réf. S3301) avec le Dako Autostainer. ÉTAPE 3 Rincer doucement à l'eau de qualité réactif puis placer dans un bain-marie pendant 2 à 5 minutes. Vérifier que tous les résidus d'hématoxyline ont été éliminés. Couvrir d'un milieu de montage permanent ou aqueux. Pour un montage aqueux, un milieu de montage de type Dako Glycergel Mounting Medium (réf. C0563), Faramount Aqueous Mounting Medium (réf. S3025) ou Ultramount (réf. S1964) est recommandé. Pour un montage permanent non aqueux, il est recommandé de sécher complètement les lames à température ambiante ou à l'air durant 1 heure à 60 ºC, puis de les incuber dans du xylène ou un substitut de xylène pendant 5 minutes. Il convient de noter que dans certains cas, des artefacts de petite taille et ponctuels peuvent apparaître sur les lames séchées à l air libre avant l'application des lamelles de protection. Pour éviter ce phénomène, il est possible de traiter les lames conformément à l'une des procédures suivantes : Option 1 : Transférer directement les lames de tissus depuis l'instrument Autostainer dans de l'alcool, en procédant soit : c) à une incubation de 2 min maximum dans de l'alcool à 99% ou d) à 10 plongées dans trois bains successifs d'alcool à 100%. Les lames doivent ensuite être plongées 10 fois dans deux bains de xylène de manière successive, avant de procéder au montage permanent. Option 2 : Transférer directement les lames de tissus depuis l'instrument Autostainer dans une série de bains d'alcool à des concentrations croissantes. Il convient de les plonger uniquement 30 secondes à chaque étape et de ne pas les exposer à du xylène. Procéder ensuite au montage permanent. Une plus longue exposition à l'alcool et/ou au xylène peut entraîner l'extraction du chromogène et ainsi l'obtention d'une coloration moins intense. Il convient de noter que ces procédures n'ont pas été testées avec tous les anticorps primaires et sont donc fournies à titre de recommandation uniquement. De même, il est nécessaire de vérifier la performance du protocole modifié de déshydratation suivi d'un montage permanent afin de garantir sa validité pour l'anticorps primaire en question. Chaque anticorps primaire utilisé doit être évalué avec le protocole modifié de déshydratation afin de confirmer que le motif de coloration est identique à celui décrit à la section "Performances" pour cet anticorps primaire. Résultats L'utilisation du tampon de substrat chromogène Liquid Permanent Red (Rouge permanent liquide) génère un produit de réaction rouge permanent sur le site de l'antigène cible ou de l'acide nucléique, qui peut être ensuite visualisé par microscopie optique standard ou microscopie fluorescente (avec des filtres Texas Red ou Rhodamine). 1 Pour une interprétation correcte, des contrôles positifs et négatifs doivent accompagner chaque nouvel échantillon. Les contrôles positifs servent à indiquer que le traitement et la manipulation des échantillons ont été effectués correctement. Les contrôles négatifs sont utiles pour évaluer une coloration non spécifique. Une activité endogène de phosphatase alcaline dans certains tissus peut générer des faux positifs. Cette activité endogène peut être inhibée par l'utilisation de Dual Endogenous Enzyme Block (réf. S2003) de Dako ou de lévamisole (réf. X3021). Le lévamisole n'inhibe pas la forme intestinale ou placentaire de la phosphatase alcaline et peut ne pas être efficace pour la coloration des échantillons contenant ces isoenzymes. Entsprechende persönliche Schutzausrüstung (PSA) tragen, um Augen- und Hautkontakt zu vermeiden. 4. Nicht verwendete Lösung ist entsprechend den örtlichen, staatlichen und EU-rechtlichen Bestimmungen zu entsorgen. 5. Auf Anfrage ist für Fachpersonal ein Sicherheitsdatenblatt erhältlich. Dako Permanentes rotes Chromogen bei 2-8 C aufbewa hren. Werden die Reagenzien nicht entsprechend den in der Produktinformation angegebenen Bedingungen aufbewahrt, müssen die Bedingungen vom Anwender geprüft werden. Nicht einfrieren. 1. Substrat-Puffer und Chromogen vor Verwendung auf Raumtemperatur abgleichen ml LPR Substrat-Puffer in ein Reagenzglas geben. Einen Tropfen LPR Chromogen hinzugeben. Gründlich mischen. 3. Das zubereitete Substrat-Puffer-Chromogen-Reagenz innerhalb von 30 Minuten verbrauchen. Um das bestmögliche Ergebnis zu erzielen, wird die Anfertigung unmittelbar vor Gebrauch empfohlen. 4. Größere Mengen der Substrat-Puffer-Chromogen-Lösung lassen sich durch Zugabe eines Tropfens Chromogen pro benötigte 3 ml Substrat-Puffer zubereiten. ( ) P04032EFG_01_K0640/ p. 5/7

6 Verfahren Nach der Inkubation mit einem mit alkalischer Phosphatase markierten Reagenz den Reagenzienüberschuss mit Wasser aus einer Waschflasche gründlich spülen (dabei den Strahl nicht direkt auf die Probe richten). Probe etwa fünf Minuten in ein Pufferbad einlegen. Nach dem Waschen überschüssigen Puffer abklopfen und Objektträger um die Probe herum vorsichtig abwischen. SCHRITT 1: SCHRITT 2: Proben mit zubereiteter LPR-Lösung bedecken Minuten inkubieren. Die optimale Inkubationszeit kann je nach Zielkonzentration und/oder Probenfixierung variieren und sollte daher durch das jeweilige Labor bestimmt werden. Mit Wasser von Reagenzqualität vorsichtig spülen. Gegebenenfalls mit Hämatoxylin gegenfärben. (Empfehlungen siehe unten.) Je nach Stärke des verwendeten Hämatoxylins bzw. der gewünschten Farbintensität der Gegenfärbung 2-5 Minuten inkubieren. Empfehlungen: Für Gewebeproben werden 2 Minuten mit Dako Hematoxylin (Code-Nr. S3002) empfohlen. Für Zytologieproben werden 4 Minuten mit Dako Hematoxylin (Code-Nr. S3309) empfohlen. Dako Automation Hematoxylin (Code-Nr. S3301) wird zur Verwendung mit dem Dako Autostainer empfohlen. SCHRITT 3: Mit Wasser von Reagenzqualität vorsichtig abspülen und 2-5 Minuten in ein Wasserbad stellen. Alle Hämatoxylinreste müssen vollständig entfernt worden sein. Mit permanentem oder wässrigem Eindeckmedium mit Deckglas versehen. Zum wässrigen Eindecken wird Eindeckmedium wie Dako Glycergel Mounting Medium (Code-Nr. C0563), Faramount Aqueous Mounting Medium (Code-Nr. S3025) oder Ultramount (Code-Nr. S1964) empfohlen. Zum nichtwässrigen, permanenten Eindecken wird empfohlen, die Objektträger vollständig bei Raumtemperatur oder bei 60 C für 1 Stunde luftzutr ocknen und dann in Xylol oder einem Xylolersatzstoff 5 Minuten lang zu inkubieren. Es ist zu beachten, dass bei Objektträgern, die vor der Anwendung von Deckgläsern an der Luft getrocknet wurden, gelegentlich kleine, punktfömige Artefakte beobachtet werden können. Dem kann mit einem der folgenden Verfahren für die Objektträger entgegengewirkt werden: Option 1: Direkter Transfer der Gewebe-Objektträger vom Autostainer auf Alkohol. Möglich sind entweder e) 99%iger Alkohol mit einer Inkubation von maximal 2 Minuten oder f) drei aufeinanderfolgende Bäder in 100%igem Alkohol mit jeweils 10-fachem Eintauchen. Danach die Objektträger 10 Mal in jeweils zwei aufeinanderfolgende Xylol-Bäder eintauchen und anschließend permanent eindecken. Option 2: Direkter Transfer von Gewebe-Objektträgern vom Autostainer in eine abgestufte Reihe von Alkoholen, aber nur für 30 Sekunden in jeder Stufe und ohne Xylol-Exposition, anschließend permanent eindecken. Ist der Objektträger länger Alkohol und/oder Xylol ausgesetzt, kann dies zu Chromogenextraktion und somit zu einer schlechteren Färbung führen. Es ist zu beachten, dass diese Verfahren nicht mit jedem primären Antikörper getestet wurden und daher nur als Richtlinien anzusehen sind. Dementsprechend muss das Ergebnis des angepassten Dehydrierungsprotokolls und des anschließenden permanenten Eindeckens überprüft werden, um sicherzustellen, dass es in Bezug auf den fraglichen primären Antikörper immer noch gültig ist. Jeder genutzte primäre Antikörper sollte mit dem angepassten Dehydrierungsprotokoll bewertet werden, um zu bestätigen, dass das Färbemuster mit dem in den Leistungseigenschaften beschriebenen Färbemuster identisch ist. Ergebnisse Permanentes rotes Substrat-Puffer-Chromogen bildet am Ort des Zielantigens oder der Nukleinsäure ein permanentes, mit einem Standard-Lichtmikroskop bzw. Fluoreszenzmikroskop darstellbares rotes Reaktionsprodukt (Texas Red oder Rhodamin-Filter verwenden). 1 Zur genauen Auswertung sollten in jedem Färbevorgang parallel zu den unbekannten Proben Positiv- und Negativkontrollen getestet werden. Positivkontrollen weisen darauf hin, dass die Probe korrekt verarbeitet und gehandhabt wurde. Negativkontrollen sind hilfreich bei der Beurteilung unspezifischer Färbungen. In einigen Gewebearten kann die endogene Aktivität der alkalischen Phosphatase zu falsch-positiven Ergebnissen führen. Diese endogene Aktivität kann durch Dako Dual Endogenous Enzyme Block (Code-Nr. S2003) oder Levamisol (Code-Nr. X3021) gehemmt werden. Levamisol hemmt weder die intestinale noch die plazentale Form der alkalischen Phosphatase und eignet sich daher möglicherweise nicht für das Anfärben von Proben, die diese Isoenzyme enthalten.
